Kurzfassung deutsch:
FT-IR spectroscopy is a versatile tool for bioprocess monitoring. In-line attenuated total reflection (ATR) probes allow for determination of media components and excreted products without the need to draw samples. Here, we used a fiber optic probe for the quantification of Penicillin V and phenoxyacetic acid, its precursor, in-line during Pencillium chrysogenum fermentations by FT-IR spectroscopy and Partial Least Squares (PLS) Regression and Multivariate Curve Resolution - Alternating Least Squares (MCR-ALS). First, the applicability of an ATR FT-IR fiber optic probe was assessed off-line by measuring standards of the analytes of interest and investigating matrix effects of the fermentation broth. Then measurements were performed inline during four fed-batch fermentations with on-line HPLC for the determination of penicillin V and phenoxyacetic acid as reference analysis. PLS and MCR-ALS models were built using these data.
