Here wereportthedevelopmentofadeviceforthetranscription-andamplification-free detectionof
DNAandRNAmoleculesdowntothezepto-molerange.Amicrofluidic chipwithabuilt-inmicroarray
wasusedformanipulationofnano-litersamplevolumes.Specific stainingandimmobilizationofthe
target molecules wasachievedviaadoublehybridizationapproachtherebyavoidingbiasduetoenzy-
matic processeslikereversetranscriptionandPCRamplification. Therefore, target molecules werein-
directlylabeledbypre-hybridizationtocomplementaryCy5-labeledprobes.Theremainingsingle-
stranded portionofeach target molecule couldsubsequentlyhybridizetocomplementary capture probes
of amicroarray.Thusa target-mediated immobilizationoflabeledDNAtookplace.Bymeansofanultra-
sensitive fluorescence readout,allmoleculeshybridizedtothemicroarraycouldbedetected.Thecom-
bination ofminimizedsamplevolumeandsinglemoleculedetectionyieldedadetectionlimitof39fM
(831moleculesin35.4nlassayvolume)for target DNA and 16fM(338molecules)for target RNA after1h
on-chip hybridization.
