English abstract:
An immense number of chemical reactions are carried out simultaneously in living cells. Nature's optimization approach encompasses the assembly of reactions in cascades and embeds them in finely tuned metabolic networks. Mimicking Nature, we established a general concept, based on the bio-retrosynthesis, for the de novo design of synthetic cascades in the model organism E. coli. These `mini pathways´ are constructed by assemble enzymes according to their functional group transformations of a particular class of substrates. Combined strategies and concepts from organic chemistry, biocatalysis, metabolic engineering and synthetic biology have been applied to develop and improve such artificial metabolic pathways.
