English abstract:
We developed a generic protocol for the stabilization of flavin-dependent monooxygenases under reaction conditions, using an engineered formulation of additives (natural cofactors and
catalytic antioxidants, Figure 1). This way, a 10^3-10^4-fold increase of the half-life was reached without resource-intensive directed evolution or structure-dependent protein engineering methods.
Furthermore, we validated our generic protocol for the stabilization of class B FMOs with three sequentially distant enzymes of this class.
