Kurzfassung englisch:
Within the different structural sugar components of glycoconjugates, one sugar family confers to the animal kingdom a special resistance to pathogens like viruses, bacteria, fungi or parasites. This sugar family is called sialic acid and is composed of a 9-carbon monosaccharide, bare a negative charge and typically occupy the distal end of glycan chain [1, 2]. The most important are the N-acetylneuraminic acids (Neu5Ac) and N-glycolylneuraminic acids (Neu5Gc). On cell surfaces of healthy human subjects glycosylation is characterized by high amounts of Neu5Ac. Upon dietary exposure to high amounts of the non-human sialic acid form Neu5Gc (predominantly found in red meats, milk products). After food consumption, Neu5Gc can be metabolically incorporated in tissues such as epithelia and endothelia [3]. If Neu5Gc is incorporated in the glycosylation instead of Neu5Ac, it produces an aberrant glycosylation, which may be the epitopes of Anti-Neu5Gc antibodies and may cause inflammatory disease, immune activation, cancer progression and immune clearance of the tissue. Immobilization of Neu5Gc on surface of polymeric support is of interest for different application in analytical chemistry and immunology for stimulating production and for determination of Anti-Neu5Gc antibodies. These glycosylated surface structures for ensure a stability must be neuraminidase resistant. Neuraminidase are a class of enzyme able hydrolyze glycosidic linkages of terminal sialic acid residue. To prevent hydrolysis by neuraminidase a carbon glycoside are used.
Briefly Neu5Ac is deacetylated and converted in Neu5Gc. The Neu5Gc is peracetylated, converted in glycosil chloride and C glycosilation is performed. Porose sulfonaed PS polymers particle are synthetized and the surface functionalized with Neu5Gc.
